HPLC PIGMENT ANALYSIS IS A COMPLICATED PROCEDURE REQUIRING EXPERT ANALYSIS AND SUBSTANTIAL COSTS FOR PURCHASING, MAINTAINING, AND CALIBRATING THE EQUIPMENT. A CENTRALIZED LABORATORY IS A COST-EFFECTIVE MECHANISM FOR PROVIDING CONSISTENTLY HIGH-QUALITY DATA TO MULTIPLE PIS AND DATABASES, BUT ONLY IF THE FACILITY VIGILANTLY PURSUES EXCELLENCE, MAINTAINS RIGOROUS QUALITY ASSURANCE AND CONTROL (QA AND QC) PROCEDURES AS PART OF A DOCUMENTED QUALITY ASSURANCE PLAN (QAP), ADHERES TO STATED PERFORMANCE METRICS TO MINIMIZE UNCERTAINTIES, AND IS EVALUATED INDEPENDENTLY (TO DETECT BIASES AND PROBLEMS). THE LATTER INCLUDES THE INTERNATIONAL SEAWIFS HPLC ANALYSIS ROUND-ROBIN EXPERIMENT (SEAHARRE), AND ANNUAL INTERCOMPARISONS OF DUPLICATE SAMPLES WITH OTHER QA LABORATORIES, INCLUDING THE PRIMARY MANUFACTURER OF STANDARDS. AN ALGAL CULTURE FACILITY WILL BE ESTABLISHED TO: A) ISOLATE STANDARDS COMMERCIALLY UNAVAILABLE OR NEEDED IN LARGE QUANTITIES, AND B) COLLECT WHOLE FILTERS TO ASSIST IN METHOD DEVELOPMENT, VALIDATION, AND QAP PROCEDURES. ALL DATA PRODUCTS WILL BE DISTRIBUTED WITH LIMIT OF DETECTION AND QUANTITATION (LOD AND LOQ, RESPECTIVELY) ANALYSES, AS WELL AS UNCERTAINTY ESTIMATES. RESEARCH ON NEW METHODS TO IMPROVE PIGMENT IDENTIFICATION AND QUANTITATION WITH THE CURRENT CVO APPROACH BASED ON THE VAN HEUKELEM AND THOMAS (2001) METHOD (VHT) WILL OCCUR IN PARALLEL WITH PRODUCTION ANALYSES. CHANGES IN THE PROCEDURES USED IN PIGMENT OCEANOGRAPHY ARE EXPECTED, ALONG WITH IMPROVEMENTS IN THE PROTOCOLS SUPPORTING NEXT-GENERATION SATELLITE ALGORITHMS, WHICH REQUIRE ADVANCED ANALYSIS TECHNIQUES AND IMPROVED PIGMENT DETECTION. ELEVATED UNCERTAINTIES ARE ASSOCIATED WITH FALSE POSITIVES AND NEGATIVES, PLUS POOR RESOLUTION BETWEEN COELUTING PAIRS. RAPID RESOLUTION LIQUID CHROMATOGRAPHY (RRLC) THE NEXT GENERATION IN LIQUID CHROMATOGRAPHY (LC) WILL BE USED TO IMPROVE THE SEPARATION OF PIGMENTS, AND DEVELOP FASTER METHODS (WHICH PROVIDES MORE ANALYST TIME FOR ALTERNATIVE DETECTION TECHNIQUES). METHOD SPECIFICA-TIONS SUCH AS THE LINEAR GRADIENT,LAG TIME, FLOW RATE, AND COLUMN TEMPERATURE WILL BE MODIFIED BASED ON DRYLAB SIMULATIONS AND ADJUSTMENTS MADE TO IMPROVE EFFICIENCY. THE DETECTION AND ANALYSIS OF PIGMENTS DIFFICULT TO DISTINGUISH BECAUSE OF SIMILARITY IN THEIR SPECTRAL ABSORPTION PROPERTIES WILL BE IMPROVED THROUGH THE USE OF LIQUID CHROMATOGRAPHY COUPLED TO MASS SPECTROMETRY DETECTION (LC-MS). THE CHEMICAL TAXONOMY (CHEMTAX) PROGRAM WILL BE USED TO EVALUATE THE IMPORTANCE OF PIGMENT DATA QUALITY ON CHEMTAX OUTPUT BY COMPARING THE OUTPUT MATRIX OF EXISTING SEAHARRE QA DATA WITH DATA FROM METHODS NOT VALIDATED WITH A PROPER QA CAPABILITY (NV). |

THIS WILL ESTABLISH THE ABILITY TO CLEARLY DISTINGUISH THE NATURE AND PARAMETERS OF THE POPULATIONS RESPONSIBLE FOR THE PRODUCTION OF THE CIB FLUCTUATIONS. 2) DETERMINE THE LYMAN BREAK, AND EPOCHS, OF THE POPULATIONS PRODUCING THESE FLUCTUATIONS BY EXPANDING THE SED OF THE MEASURED FLUCTUATIONS. 3) CROSS-CORRELATE THE MEASURED FLUCTUATIONS WITH OPTICAL, IR, X-RAY, AND MICROWAVE IMAGING FROM A NUMBER OF OTHER FACILITIES. THESE INCLUDE BOTH THE SPITZER/IRAC AND HST/WFC3+ACS INSTRUMENTS, TO HELP CHARACTERIZE STELLAR (NUCLEOSYNTHETIC) SOURCES OF THE CIB FLUCTUATIONS. EFFORTS ALSO INCLUDE CROSS-CORRELATING WITH IMAGING FROM COEXTENSIVE X-RAY OBSERVED FIELDS, IN ORDER TO MEASURE THE CONTRIBUTION OF BLACK HOLES (AGN AND OTHER GRAVITATION OR ACCRETION-POWERED SOURCES) AMONG THESE SOURCES TO THE X-RAY BACKGROUND. CROSS-CORRELATING THE NEAR-INFRARED EUCLID CIB MAPS WITH PLANCK SUB-MM CIB AND CMB MEASUREMENTS WILL PROBE THE DUST EMISSION FROM THESE SOURCES AND CONSTRAIN ANY THERMAL SZ COMPONENT CONTRIBUTION. LIBRAE WILL THUS IDENTIFY THE NET EMISSIONS FROM THE FIRST STARS ERA, LEAD TO A BETTER UNDERSTANDING THE CONDITION OF INTERGALACTIC MEDIUM AT THAT EPOCH, ISOLATE THE CONTRIBUTIONS FROM THE FIRST BLACK HOLES AND SHED LIGHT ON DUST CONTENT AT THOSE TIMES. THE PROJECT HAS POTENTIALLY TRANSFORMATIVE IMPLICATIONS FOR UNDERSTANDING THE EMERGENCE OF THE UNIVERSE OUT OF THE "DARK AGES". IT IS WELL ALIGNED WITH EUCLID MISSION OBJECTIVES. |
